PERSONAL STATEMENT

I decided to pursue the PhD in medicinal chemistry for two major reasons: firstly, I am fascinated with the process of drug discovery and synthesis, and secondly, I wish to help find a way to integrate Traditional Chinese Medicine (TCM) with modern science.

The experiences of recoveries from common cold and high fever when I was a kid by drinking cups of “medicated soup” decocted from more than ten kinds of herbs prescribed by an old TCM doctor with professional inheritance of many generations have made deep and long lasting impression on me.

TCM is characterized by its concept of holism, the principle of syndrome identification in clinical diagnosis/treatment, its herbal compound medication in pharmacotherapy and its curative effect demonstrated in medical practices over thousands of years in China. However, its basic theory is lacking scientific proof, and the mechanism of actions of herbal components is yet to be clarified by modern science and technology. Medicinal Chemistry emphasizes on cultivating skills and teaching relevant knowledge in the area of drug discovery/synthesis and identification of mechanism of actions of novel compounds, so I feel that by taking this program, I will be able to participate in the discovery of mechanism of actions of Chinese herbal medicines and the clarification of meanings of TCM theory.

My undergraduate study in Biochemistry in the University of XX helped me laid firm foundation in fundamentals for advanced studies in medicinal chemistry. Many courses I took during the undergrad study had lab components, yet I enjoyed the organic labs the most because firstly, it was fascinating to react different chemicals with one another in specific orders and observe the formation of new compound; secondly, different from some other labs which require me to have lab partners, the organic lab instructor encouraged us to work independently. At first I got really stressed up because there was a time limit to finish each experiment and I was not sure whether I could get everything done correctly on my own; yet after a few sessions, I got used to and started to enjoy working and thinking independently and self confidence also started to build up. Another experience that served to consolidate my determination in pursuing medicinal chemistry is that during my undergraduate study, I took a course named Environmental and Natural Products Biochemistry which not only gave me general information about the functions, distributions and environmental actions of a variety of natural compounds isolated from plants and other organisms, but also helped me discover where my true interest lies. However, there was no laboratory associated with that course in our university, and as a result, I didn’t manage to find a chance to try my hands on synthesizing those natural compounds. I have been looking forward to participate in researches related to drug discovery/synthesis since then.

My graduate study in Pharmaceutical Sciences in XX University not only provided me with more detailed knowledge in drug discovery, but also helped me develop valuable research skills which will help me succeed in the medicinal chemistry program. During my study in this university, I participated in a research project concerned about improving physicochemical properties of a poorly soluble carboxylic acid drug (gemfibrozil) by salt formation, which generally involved synthesizing a series of amine salts of gemfibrozil, comparing various properties (include saturated solubility, IDR, melting points by DSC, particle size, surface tension and crystal habits) of the resulting salt forms and exploring possible trends existed among these properties (at the end of the project I successfully discovered a negative correlation between log salt solubility and salt melting points; positive correlations between log salt solubility and pHmax/log IDR; and a negative correlation between number of rings presented in the counterions and the solubility of the corresponding salt forms of gemfibrozil). Although this project is not directly related to medicinal chemistry, it helped to greatly enhance my research skills, which include doing literature review on unfamiliar concept, then quickly internalizing these new concepts and applying them onto the experiments that I was doing; making realistic work plans; effectively managing my time during each lab session and accomplishing every measurement as accurately as I could to avoid repeating experiment which would put me under the risk of falling behind my schedule. The project report writing improved my ability to critically analyze and compare literature findings and draw reasonable conclusions from my own data based on the knowledge obtained from published papers.

The undergraduate and graduate study experiences have tempered me as a self-reliant person who is always ready to approach problems independently with confidence; cultivated my interest in drug discovery/synthesis; and helped me develop research skills that are essential for success in medicinal chemistry program. ……此处为申请学校及教授信息
